Donald Kohn iz Eli i Edythe Centra za regenerativnu medicinu i istra\u017eivanje o mati\u010dnim \u0107elijama na UCLA, svih devet beba su izlije\u010dene. 10 ispitanik je tinejd\u017eer koji u trenutku ispitivanja nije imao naznake za pobolj\u0161anjem imunog sistema. Kohnov metod je genska terapija mati\u010dnim \u0107elijama koja sigurno pobolj\u0161ava imuni sistem kod beba sa imunodeficijencijom koriste\u0107i njihove mati\u010dne \u0107elije.<\/p>\n<p>Adenozin-deaminazna deficijencija, kombinovana sa imunodeficijencijom, tako\u0111er poznata i kao ADA-SCID ili te\u0161ka kombinovana imunodeficijencija, je uzrokovana genskom mutacijom adenozin-deaminaza enzima, koji je va\u017ena komponenta imunog sistema. Bez ovog enzima, \u0107elije imunog sistema nisu u stanju da se odbrane od infekcije. Djeca sa ovom bolesti moraju biti izolovana potpuno i u \u010distom prostoru da bi se izbjeglo izlaganje virusima i baterijama, \u010dak i ako im je hladno to mo\u017ee biti fatalno po njih.<\/p>\n<p>Trenutno, postoje dva tretmana za ije\u010denje djece sa ADA-SCID. Mo\u017ee im se davati dva puta sedmi\u010dno injekcija adenozin-deaminaza enzima. To je cjelo\u017eivotni proces koji mo\u017ee biti veoma skup a u ve\u0107ini slu\u010dajeva ne\u0107e vratiti imuni sistem do optimalnog nivoa. Neka djeca budu podvrgnuta transplantaciji ko\u0161tane sr\u017ei, od odgovaraju\u0107eg donora, naj\u010de\u0161\u0107e bliskog srodnika, ali kod transplantacije ko\u0161tane sr\u017ei mo\u017ee do\u0107i do odbijanja transplantiranih \u0107elija donora.<\/p>\n<p>Istra\u017eiva\u010di su koristili strategiju koja ispravlja ADA-SCID mutaciju pomo\u0107u njihovih geneti\u010dki modifikovanih mati\u010dnih \u0107elija, koje mogu stvarati sve tipove krvnih \u0107elija. Mati\u010dne \u0107elije svakog djeteta su analizirane u laboratoriju i u njih se insertovao odgovaraju\u0107i gen za stvaranje adenozin-deaminaza enzima. Svako dijete je primilo terapiju sa vlastitim mati\u010dnim \u0107elijama u koje je insertovan gen.<\/p>\n<p>Klini\u010dko ispitivanje je trajalo od 2009 do 2012 godine i tretirano je 10-ero djece sa ADA-SCID koja nisu imala odgovaraju\u0107eg donora ko\u0161tane sr\u017ei. Troje djece je tretirano u Nacionalnom institutu za zdravlje a ostalih sedam ispitanika je tretirano i lije\u010deno na UCLA. Nijedno dijete nije pokazivalo komplikacije nakon tretmana. Devet od deset ispitanika sada imaju dobar imuni sistem i ne moraju vi\u0161e biti izolovani. Ta djeca \u017eive sasvim normalan \u017eivot, igraju se u prirodi, idu u \u0161kolu, ali \u0161to je i najva\u017enije mogu da se izlije\u010de od prehlade ili od infekcije uha. Tinejd\u017eer koji nije bio izlije\u010den i dalje prima enzimsku terapiju.<\/p>\n<p>\u010cinjenica da je 9 beba izlije\u010deno a tinejd\u017eer nije, ukazuje na to da genska terapija za ADA-SCID djeluje bolje kod mla\u0111ih pacijenata, prije nego njihova tijela izgube mogu\u0107nost vra\u0107anja imunog sistema.<\/p>\n<p>Sljede\u0107i korak je da se tra\u017ei odobrenje od Uprave za hranu i lijekove (FDA) za gensku terapiju s nadom da \u0107e ovaj tretman pomo\u0107i svakom djetetu sa ADA-SCID bolesti. Kohn i saradnici su tako\u0111er prilagodili terapiju mati\u010dnim \u0107elijama djeci sa anemijom srpastih \u0107elija i X-vezanu hroni\u010dnu granulomatoznu bolest, koja se uobi\u010dajeno naziva X-vezana CGD.
